According to BN, the update is as follows:

Software Updates - Version 1.0.1
Barnes & Noble is providing a minor update to NOOKcolor firmware that will improve NOOKcolor's Wi-Fi connectivity and browser experience (such as being able to easily toggle between desktop and mobile mode).

The update is available from BN's software update page.

The download is a signed 161 meg zip file, and will not install on rooted Nooks. A special page for helping people who have rooted is available here.

Warning: Let us say that again. The update will not install on a rooted NC. You must restore the /system folder to factory by re-flashing it or you can not install the update.

The update is a complete replacement of the /system file system of the NC. It is not possible for rooting to survive the process, and rooting will have to be re-done.

When the process is complete, the installation records of your installed software other than the factory apps are also deleted. The use of a program like Titanium Backup is strongly urged to restore apps and their settings after updating and re-rooting.

A complete file system diff of the update and the 1.0.0 version is available here.
